Purpose and Background:

The offer of this lab is to show the fit way of preparing a civilisation to insure that no contaminating beingnesss argon introduced into culture materials when they are inoculated or handled in some manner. It also insures that organisms that are world handled do not contaminate the handler or others who whitethorn be present and that no contamination remains later the work has been completed.

Hypothesis

The theory illustrated by this laboratory experiment is that if proper Aseptic techniques are followed contamination will not occur.

Materials employ in this laboratory experiment.

·Broth culture of E. coli
·Tubes of sterile food for thought descent
·Inoculating tat
· bunsen burner
·disinfectant

Procedure

1.Disinfect the work surface by spraying it down with a disinfectant and then disinfect with paper towel
2.Light Bunsen burner
3. sterilize inoculating closed circuit by holding it over the flame of the Bunsen burner until the circus tent becomes bright red.
4.Using free hand, gently shake the subway of E. coli culture broth to disperse the culture throughout the broth.
